The correct classification and identification of Euphorbia plant species is an essential point in the study of these important plants used for ornamental purposes. The main objectives of this study were to evaluate the protein content and antigens of eight leaf samples of E. macrosulcata, collected from Argentina, and to examine the relationship between leaf characteristics and chemical composition (fat, protein, carbohydrate, ash, dietary fiber, and sodium content) by principal component analysis. In addition, to identify antigens from E. macrosulcata, we followed a protocol that involves enzyme-linked immunosorbent assay (ELISA) and western blotting. In addition, the woody stems, leaves, and fruits of E. macrosulcata were tested for mining antigens by IEF. Four leaf samples showed nitrogen-to-phosphorus ratio greater than 4.0, suggesting nitrogen excess. Results showed a positive correlation between total protein content, and leucine/isoleucine ratio. The addition of galls is a common practice in Argentina. Approximately 5 g of galls can be obtained from 1 kg of leaves. We detected mining antigens (22, 24, and 26 kDa) and a N-terminal sequence of 26 kDa in all leaves. Our results show the importance of antigenic and chemical studies for a complete identification of the E. macrosulcata species.
